Pulmonary toxicity with mefloquine. E. Udry, F. Bailly, M. Dusmet, P. Schnyder, R. Lemoine, J.W. Fitting. #ERS Journals Ltd 2001. ABSTRACT: This report presents a case of acute lung injury developing within hours after administration of mefloquine for a low-level Plasmodium falciparum malaria, which was persistent despite halofantrine therapy. Extensive microbiological investigation remained negative and video-assisted thoracoscopic lung biopsy demonstrated diffuse alveolar damage. The evolution was favourable without treatment. This is the second report of acute lung injury and diffuse alveolar damage caused by mefloquine. Glucose-6-phosphate dehydrogenase deficiency was present in the former case and was thought to contribute to the lung injury. However, glucose-phosphate dehydrogenase was normal in the present case, suggesting that it is not a predisposing condition to the lung injury. Mefloquine-associated dizziness, diplopia, and central serous chorioretinopathy: a case report
BACKGROUND Many acute and chronic neurological sequelae from the quinoline derivative antimalarial drug mefloquine, including dizziness and effects on the visual system such as diplopia and blurred vision, may be attributable to focal central nervous system toxicity. Maculopathy has also been reported with use of mefloquine, although the mechanism of this effect has remained unclear. Identifica...
متن کاملLong-term Follow-up Study of Pulmonary Function Test in Children with History of Hydrocarbon Aspiration
Background Aspiration of hydrocarbons causes several acute and chronic pulmonary complications; it may even lead to death. The aim of this study was to investigate the types of long-term pulmonary complications in children with a history of hydrocarbon aspiration. Materials and Methods In this case-control study, 21 children ...
متن کاملInhibition of volume-regulated and calcium-activated chloride channels by the antimalarial mefloquine.
We have used the whole-cell patch-clamp technique to study the effect of mefloquine (Lariam), a commonly used antimalarial drug, on the volume-regulated anion channel (VRAC) in cultured bovine pulmonary artery endothelial cells. We also examined its effects on other Cl(-) channels, i.e., the Ca(2+)-activated Cl(-) channel and the cystic fibrosis transmembrane conductance regulator, to assess th...
متن کاملIdiosyncratic quinoline central nervous system toxicity: Historical insights into the chronic neurological sequelae of mefloquine☆
Mefloquine is a quinoline derivative antimalarial which demonstrates promise for the treatment of schistosomiasis. Traditionally employed in prophylaxis and treatment of chloroquine-resistant Plasmodium falciparum malaria, recent changes to the approved European and U.S. product labeling for mefloquine now warn of a risk of permanent and irreversible neurological sequelae including vertigo, los...
متن کاملMefloquine use, psychosis, and violence: A retinoid toxicity hypothesis
Mefloquine use has been linked to severe gastrointestinal and neuropsychiatric adverse effects, including cognitive disturbances, anxiety, depression, psychosis, and violence. The adverse effects of the drug are thought to result from the secondary consequences of hepatocellular injury; in fact, mefloquine is known to cause a transient, anicteric chemical hepatitis. However, the mechanism of me...
